AB OBJECTIVE: To study lymphomagenesis associated with SIV/HIV-2 infection
in cynomolgus monkeys (Macaca fascicularis). METHODS: Monkeys were
infected with SIVsm or HIV-2 and were studied for clinical signs of
immunodeficiency and consecutive biopsies examined for B-cell clones and
association with herpes virus, by immunohistopathology, PCR, Southern
blot (SB), in situ hybridization, electron microscopy and cell culture.
RESULTS, DISCUSSION AND CONCLUSIONS. Monkeys infected with SIVsm but not
these with HIV-2 developed frequently (21/51) malignant lymphomas.
Lymphoma induction time varied from 5 to 46 months. All monkeys with
lymphoma were markedly immunosuppressed. Most lymphomas were primarily
extranodal including CNS, of high-grade malignant histology, of B-cell
origin and associated with a B-lymphotropic herpes virus related to
human EBV (HVMF). HVMF showed considerable restriction map homology with
EBV in the EBNA-1,2 regions and within the IR1, the EBER and the ori-P
regions, but not with LMP regions. HVMF from a monkey lymphoma cell line
transformed in vitro normal monkey B-cells. By in situ hybridization for
EBER-RNA, HMVF associated lymphoma cells could be detected before
clinical lymphoma manifestation. This was also indicated by the early
appearance of a dominant clone according to DNA-PAGE of the VDJ
immunoglobulin heavy chain region. These observations indicate a marked
similarity of lymphomas in SIV immunosuppressed monkeys with human
AIDS-lymphomas associated with EBV and the possibility to use this
monkey lymphoma model for evaluation of therapeutic protocols.
